A general method is described for large scale preparation of 13C and 15N region-specific enriched DNA oligomers that can be used for NMR studies. The procedure combines two enzymatic polymerisation reactions: first of 13C,15N-labeled and then of nonlabeled deoxyribonucleotides. It is applied to the regional labeling of one 16mer strand of a 64 residue four-arm DNA structure that serves as a model for the Holliday junction intermediate in genetic recombination and repair.
