• S219V mutant has been shown to be more stable and more efficient as a catalyst than the wild-type protease
• pETTev produces an MBP-His-TEV fusion protein with an internal cleavage site
• The product self-cleaves intracellularly to yield His-tagged protease, MW = 28.5 kDa
