Calmodulin Mutant: F92A
Engineered mutant of isoform 1 calmodulin, from Homo sapiens (InfoCard)
General Effects of the Mutation:
- This mutation is sufficient to cause a temperature-sensitive phenotype in yeast (InfoCard)
- This mutation causes a decrease in the thermal stability of the isolated Cam-C domain, as indicated by a 11.8 degree Celsius shift in the far-UV CD thermal dentauration curve. It also increases the calcium affinity. The calcium-dependent increase in bisANS fluorescence is 45% less in CaM-Ca with the F92A mutation than in wildtype CaM-C. This fluorescence increase monitors how hydrophobic the environment of the probe is-- a larger increase corresponds to a more hydrophobic environment (InfoCard)
