MALDI Protocol
Desalted Samples
Solution A: .1% Tryfluoroacetic acidm; 1 ul TFA in 1mL water
Solution B: 6:4 acetonitrile water; 60 uL : 40 uL
1. Wash tip with 2x 10 uL of B
2. Wash tip with 2x 10 uL of A
3. Bind 10ul protein to column 15 times
4. Wash with 3x 10 uL of A
5. Elute with 5uL of B 15 times.
Use Zip Tips (Millipore cat # ZTC 185096)
1. Reserve Mass Spec, fill out form and clean off target with MetOH.
2. Prepare 2uL desaulted sample plus 2uL matrix (sinapinic acid)
3. Add sample to MALDI target, avoid circular depressions because it's easier to clean.
4. Allow sample to dry. It should turn white if there is enough matrix.
5. Turn on light.
6. Open user file and select "Eject" Load target and select "Load"
7. Use method : "LDE1001" for proteins
8. Set calibration: Default
9. Ctrl-Pgup increases Laser Power. Increase to ~2000 (more or less by ~200 or until peak is observed)
10. Hit Joystick tip button when laser is set and perform two-point calibration by hitting button B. and following on screen instuctions
11. Proceed to next sample
